Correlations between Neuropsychological and Neuroimaging Data in Myotonic Dystrophy

چکیده
Twentyfour patients with Mytonic Dystrophy (MD) (mean age 42.8 ±13.4 yrs; range 21-59) of moderate degree underwent neuropsychological and neuroradiological (Magnetic Resonance Imaging MRI) evaluation. The results show the presence of cognitive impairment, in particular involving visuo-constructive and attentional skills, not related to sex, age at onset or disease duration; subjects with maternal inheritance presented however a poorer performance in respect to those with paternal inheritance. On MRI scans subcortical white matter hyperintense lesions (WMHL) have been found in 18/24 cases, which did not correlated with both the clinical variables and the cognitive impairment. These lesions, in particular those located in the temporal lobes (6/24 subjects), seem to be characteristic of the disease and are probably scarcely progressive.

Genotype–Phenotype Correlations in Iranian Myotonic Dystrophy Type I Patients
Objectives: Myotonic Dystrophy type I (DM1) is a dominantly inherited disorder with a multisystemic pattern affecting skeletal muscle, heart, eye, endocrine and central nervous system. DM1 is associated with the expansion and instability of CTG repeat in the 3chr('39') untranslated region of the myotonic dystrophy protein kinase (DMPK) gene located on chromosome 19q13.3. The aim of this study w...

Myotonic dystrophy types 1 (DM1) and 2 (DM2) are similar yet distinct autosomal-dominant disorders characterized by muscle weakness, myotonia, cataracts, and multiple organ involvement, including the brain. One key difference between DM1 and DM2 is that a congenital form has been described for DM1 only. Expression of RNA transcripts containing pathogenic repeat lengths produces defects in alter...
متن کاملThe brain in myotonic dystrophy 1 and 2: evidence for a predominant white matter disease
Myotonic dystrophy types 1 and 2 are progressive multisystemic disorders with potential brain involvement. We compared 22 myotonic dystrophy type 1 and 22 myotonic dystrophy type 2 clinically and neuropsychologically well-characterized patients and a corresponding healthy control group using structural brain magnetic resonance imaging at 3 T (T(1)/T(2)/diffusion-weighted). Voxel-based morphomet...
متن کاملCharacterization of the pattern of cognitive impairment in myotonic dystrophy type 1.
BACKGROUND Central nervous system involvement occurs in most patients with myotonic dystrophy type 1 (DM1): mental retardation characterizes congenital forms, while a mild cognitive impairment has been described in adult patients with classic DM1. Neuropathological studies documented neurofibrillary tangles and an aberrant tau-protein expression in brain tissues of patients and animal models of...
متن کاملRelationship between neuropsychological impairment and grey and white matter changes in adult-onset myotonic dystrophy type 1
Myotonic dystrophy type 1 (DM1) has a wide phenotypic spectrum and potentially may affect central nervous system with mild to severe involvement. Our aim was to investigate grey matter (GM) and white matter (WM) structural alterations in a sample of adult-onset DM1 patients and to evaluate relationship with clinical and cognitive variables. Thirty DM1 patients underwent neuropsychological inves...
